چکیده

In industrial hydrolysis processes the lignocellulose-degrading enzymes may have a potential application as an alternative to chemical treatments because they produce high specificity, mild reaction conditions, no toxic chemical hydrolysis inhibitors, and no loss of substrate due to chemical modifications. The amounts of reducing sugars released from the degradation of wheat straw by the action of endoxylanase, peroxidase, β-xylosidase, and α-Larabinofuranosidase, which were produced by T. fusca BD25, were equal to 3.0%, 0.2%, 0.3%, and 0.2% hydrolysis (as xylose equivalents) of the substrate used, respectively, after 10 h of incubation. In the same conditions, the hydrolysis rate of endoglucanase, which was a commercial preparation from Trichoderma viride was 4.9%. However, in order, addition of peroxidase, endoglucanase, β-xylosidase, and α-L-arabinofuranosidase preparations to the endoxylanase preparation significantly enhanced the reducing sugar yields (1.33-, 1.52-, 1.61-, and 1.71-fold, respectively) and the hydrolysis of wheat straw (4.3%, 12.3%, 13.5%, and 14.7%, respectively). The concentrations of phenolic-compounds as ligninequivalents released from wheat straw by the actions of lignocellulose-degrading enzyme preparations either acting alone or in combinations ranged from 2.5 to 129.3 μg mL (as p-coumaric acid equivalents). The combination of purified lignocellulose-degrading enzymes significantly increased the degradation of straw 1.33to 1.71-fold. The results highlight the role of lignocellulose-degrading enzymes in the degradation of wheat straw and suggest that the use of enzyme cocktails may significantly improve the hydrolysis of wheat straw in industrial processes.

Sacchariiication of Straw by Actinomycete Enzymes

Over 200 strains of actinomycetes, representing nine distinct genera, were screened directly for the ability to release reducing sugar from ball-milled wheat straw, using a microtitre plate assay system. Xylanase activity was detected in nearly all of the strains examined while activities against purified cellulosic substrates were less widespread and relatively low. Straw saccharification resu...

Differential proteomic analysis of the secretome of Irpex lacteus and other white-rot fungi during wheat straw pretreatment

BACKGROUND Identifying new high-performance enzymes or enzyme complexes to enhance biomass degradation is the key for the development of cost-effective processes for ethanol production. Irpex lacteus is an efficient microorganism for wheat straw pretreatment, yielding easily hydrolysable products with high sugar content. Thus, this fungus was selected to investigate the enzymatic system involve...

Assembly of Xylanases into Designer Cellulosomes Promotes Efficient Hydrolysis of the Xylan Component of a Natural Recalcitrant Cellulosic Substrate

UNLABELLED In nature, the complex composition and structure of the plant cell wall pose a barrier to enzymatic degradation. Nevertheless, some anaerobic bacteria have evolved for this purpose an intriguing, highly efficient multienzyme complex, the cellulosome, which contains numerous cellulases and hemicellulases. The rod-like cellulose component of the plant cell wall is embedded in a colloid...

Characterization of β-Xylosidase and α-L-Arabinofuranosidase Activities From Thermomonospora Fusca BD25

Thermomonospora fusca BD25 produces relatively high levels of activity of β-xylosidase and α-L-arabinofuranosidase. The aim of the work described in this study was to characterize some properties of β-xylosidase and α-L-arabinofuranosidase produced by T. fusca BD25 when growing on oat spelt xylan as the main carbon and energy sources. The substrates p-NPX and p-NPA reacted with β-xylosidase and...

Uncovering the Potential of Termite Gut Microbiome for Lignocellulose Bioconversion in Anaerobic Batch Bioreactors

Termites are xylophages, being able to digest a wide variety of lignocellulosic biomass including wood with high lignin content. This ability to feed on recalcitrant plant material is the result of complex symbiotic relationships, which involve termite-specific gut microbiomes. Therefore, these represent a potential source of microorganisms for the bioconversion of lignocellulose in bioprocesse...
